Functions & Requirements

Functions Requirements
Network of closed loop channels
  • Rectangular shape
  • Polydimethylsiloxane (PDMS)
  • diameter of channels: 100nm –  900μm

Multiple in vitro model application

  • PDMS allows for numerous biological applications:
    • CTCs
    • Hematologic Diseases
    • Polymerase Chain Reactions

  • Chemical signal isolation

  • Plasmolyzed PDMS to prevent signal molecule absorption

Device platform does not degrade chemical signaling molecules

  • PDMS is organic biocompatible
    • it is non-toxic
    • low chemical reactivity

Endothelialization of the channels
  • Monolayer of endothelial cells can adhere to device channels
  • Fibronectin concentration of 2µg/cm2
Imageable for long term studies

  • PDMS is transparent
  • Use of small heating elements and element placement do not obstruct view of channels

Thermal convection
  • Use of integrated Nichrome wire to heat the device, and  create temperature gradient
  • Create the buoyancy flow
  • PDMS thermal conductivity (0.15 W/m K)

Controlling Temperature Gradient
  • Thermocouple
  • Controller
  • Temperature accuracy of ± 0.5 °C
Device should be no bigger than 1inch x 3 inch in total

  • Device should fit on standard 1 x 3 inch glass slide
  • Has to fit in imaging device

Flow velocity
  • 1cm/minute
